You're on the right track but James the 1st was a Stuart not a Tudor

"Roanoke Island was the site of the 16th century Roanoke Colony, the first English colony in the New World in what was then called Virginia, in honor of England's ruling monarch, Queen Elizabeth I." There were two failed attempts to establish a permanent colony on Roanoke Island.

Perhaps the question would be better stated as: What is the first American colony named after a Tudor Monarch. I read the quetion as getting the first American colony and then finding out after whom it was named. However, I admit that I did not realize that James I was a Stuart.
Good question nontheless.

You are right Goobas. Until you responded I didn't think there would be any ambiguity.
Virginia itself is mentioned as the first 'colony' in the site I was on and others did back this up.
On furthur reference both are mentioned in varying degrees but Elizabeth I is the only Tudor Monarch, She is the last Tudor and James I followed her as the first Stuart so it was all happening in both their reigns
"1584 - Sir Walter Raleigh founds the first American colony and names it Virginia after Elizabeth the Virgin Queen"
