Maurice Elvey was a good example of a prolific director, having made over 100 films, but as his career stretches back into the silent era I imagine most of them are only shorts:

Satoru Kobayashi is a Japanese director who has pumped out over 400 feature length pink films, that is of the soft porn variety:

In the realm of Hollywood feature films, William Beaudine is credited with the most, at 182. Richard Thorpe, who trails by 3 with 179, directed more A and B list films, less serials and explotation films than Beaudine, so he would have to be considered as well.

Spanish director Jesus Franco also has over 180 films, almost all low budget explotation slasher pics.
People such as D.W. Griffith, Mack Sennett, Alice Guy, Georges Méliès or Allan Dwan directed hundreds and hundreds of films, but most were 1 or 2 reelers and many did not officially credit them for their work.
Sennett alone has producer credit on over 1000 films.
