The first character to speak in Star Wars (Episode IV, A New Hope) is C3PO. I believe he says, 'What was that?'. My source is my memory. Huge Star Wars fan here!

The first character to speak in Star Wars was C-3PO. He says, "Did you hear that R-2? They've shut down the main reactor. We're doomed. There will be no escape for the princess this time."

Before R-2 replys with beeps or whistles, C-3PO says as typed below.

C-3PO: Did you hear that? (Asking R-2) They've shut down the main reactor. We'll be destroyed for sure. This is madness!" There is a pause as Rebel troopers take positions along the Tantive IV's corridor. Then C-3PO replies, "We're doomed!"
