LSD was originally developed as a truth serum, and in the right doses, it seems to work. In early trials, it was given to a young girl and she kept talking for something like 18 hours without a break. They reckoned she had narrated every single thing that had happened to her in her short life.

Sodium Pentathol has sometimes been called a 'truth serum.' It seems to inhibit some higher brain functions that make you think it's necessary to lie. Beer does this too. Anything that releases inhibitions probably has this effect in some way. Of course, the brain is a very complex thing, and the concept of 'truth' is so fuzzy, that it's probably impossible to have a drug that reliably makes anyone tell the truth.
