    Which actually means between four and six were murdered (depending on your opinion of Lady Jane Grey and whether regicide (Charles I) is murder or not).

    Two English kings were murdered by opponents, after they had been deposed. The first was Edward II who earlier in his reign had lost the vast tracts of Scotland that his father Edward I had acquired. Many of the nobility plotted against Edward II (including his own wife, Queen Isabella), and although he put down one rebellion he was rapidly losing the plot. In 1326 a second rebellion forced him to flee westwards, and he was deposed in January 1327 to be succeeded by his son Edward III (who had sided with his Isabella). Later that year Edward II was horribly murdered at Berkeley Castle, Gloucestershire.

    The second was Henry VI, who in a similar way lost vast acreages in France. He was prone to bouts of insanity and during these Richard, Duke of York was appointed Protector to rule in his place. This caused massive rifts of loyalty and the Wars of the Roses broke out as Henry and Richard slogged it out. When Richard was killed, his son Edward took up the cause. The Yorkists eventually won and Henry went into exile whilst Edward became Edward IV. In 1471 Henry made his comeback and it was the turn of Edward to flee, but the comeback could not be sustained. Later that year, Edward got his throne back, and Henry found himself in the Tower of London. This time, Edward made sure that there was to be no further comeback - he had Henry stabbed to death.

    I thought that Britain did not exist until 1603 so it would be better to add on some Scottish ones.

    I'm afraid Scots kings were far more likely to be murdered than their English counterparts. Family feuding resulted in the murder of Duncan I by Macbeth's family and the deaths of Macbeth and his son Lulach at the behest of Ducan's son Malcolm III. Donald III, Malcolm's younger brother was overthrown by Malcolm's son who became Duncan II, but Donald murdered Ducan to regain the throne. Malcolm's eldest son by his second marriage then intervened by overthrowing Donald, blinding him, and holding him in prison in such harsh circumstances that he died very quickly.

    After all this eleventh century violence, the Scots behaved themselves until 1437 when their reforming king, James I was set upon in Perth by a band of discontents and murdered.

    Richard II of England may also be a candidate. I don't think we know exactly how he died in Pontefract Castle, but he's widely thought to have been murdered (possibly starved to death) by his captors.
