They left Southampton, but the other ship the Speedwell sprang a leak, so they put into Plymouth where most of her passengers were transferred to the Mayflower. Yes, I'm right!!!

So the answer is Plymouth AND Southampton !
Those originally on the Mayflower set sail from Southampton, those originally on the Speedwell also set sail from Southampton but NOT on the Mayflower. When they set sail on the MAYFLOWER it was from Plymouth.

Whoops I pasted the additional info in the wrong bit.
According to http://www.pubs.com/mayfse16.htm the journey started from London. From a Rotherhithe quayside, near a pub called the Shippe, the Mayflower set sail for America. It was the spring of 1620, on board were a group of Protestants fleeing religious persecution.
Captain Christopher Jones couldn't have known that his passengers were to become the most famous Americans ever, the Pilgrim Fathers.
The Mayflower and its crew returned to Rotherhithe in 1621. Jones died a year later and was buried at St. Mary's Churchyard, a stones throw from the pub. His grave is now unmarked but a plaque records his fateful journey.
A century later the Shippe was rebuilt and renamed the Spread Eagle and Crown. In 1957 the pub was restored.

Mark's first answer was technically correct as the ship would have had to have the sails set when it left Plymouth. The question should have added the word 'first' to make Plymouth wrong.

If you want to be totally pedantic it was only 1 person who came from Rotherhithe. The journey actually started from Holland where in 1608 a group of English Puritans fled to escape persecution for their beliefs - but finding it impossible to settle there, they sought and obtained permission to emigrate to the English colony of Virginia.
In July 1620 they sailed to England in the Speedwell, to join up with another group of Puritans who were to sail in the Mayflower. The two ships left Southampton together, but the Speedwell sprang a leak, and then put into Plymouth, where most of her passengers were transferred to the 180-ton Mayflower.

Plymouth was never on the route from SOUTHAMPTON to America,they only put in there as they had to turn back owing to the damage to the Speedwell.Pedantic Siskin is 100 per cent correct
