In the year 2554 it was the beginning of Captain Christopher Pike's first five-year mission aboard USS Enterprise NCC-1701

But there was a previous starship to the Enterprise because in episode 2 "Where No Man Has Gone Before", near the edge of the galaxy, they discover a 200-year-old ship recorder from the USS Valiant. So Enterprise cannot be the name of the "original" starship.

Also in either The Cage or the Menagerie (can't remember which) when Captain Pike and Dr. Boyce, the ship's surgeon are having a martini, Pike mentions his predecessor, Captain Robert M. April.
I found this download but I haven't looked at it. Thought I'd post it for any Star Trek fans. It's Gene Roddenberry's original Star Trek NBC story outline.

http://www.mts.net/~arphaxad/chronost.html has the chronological sequence and mentions Captain Robert M. April. You could argue that in one of the episdodes Kirk goes back in time to Earth in 1930 so he was the first. If you ignore Earth, the earliest date was 2729 BC- "All Our Yesterdays" which also had James T. Kirk.
