William Shatner in the episode 'The Enemy Within'.
When a transporter accident splits Captain James T. Kirk (William Shatner) into two selves (the passive and the aggressive), chaos ensues. The inevitable Kirk vs. Kirk showdown is the first in what would be a recurring theme for the Enterprise captain. Far and away, Shatner leads the pack in terms of sharing screen time with himself, having engaged in hand-to-hand combat with his double, duplicate or doppelganger on multiple occasions.

Mark Lenard is one of the few actors ever to portray three different aliens on Star Trek: a Romulan, a Klingon and a Vulcan.
He played the Romulan commander in the original series' episode "Balance of Terror," and the Vulcan Sarek (Spock's father) in the episode "Journey to Babel".
In Star Trek - The Motion Picture, he played the un-named Klingon captain.
He also almost played a human -- Gene Roddenberry wanted him to play Abraham Lincoln in "The Savage Curtain" (the episode where Kirk, Spock, Lincoln, and Vulcan legend Surak represent "good" in a staged battle with Genghis Khan and three others representing "evil"), but Lenard was unavailable at the time.
In addition he played the first Klingon to feature a "forehead" (the lumpy forehead prosthetics).

Diana Muldaur plays Dr. Katherine Pulaski in TNG.
She also played "Dr. Miranda Jones" in the episode "Is There in Truth No Beauty?" of the original series.
In addition she also portrayed "Lt.Cmdr.Ann Mulhall, Ph.D" and "Thalassa" in the episode "Return to Tomorrow"

Majel is the only person to appear in all Star Trek series, even if only as a computer voice. In the pilot episode "The Cage" she played Number One, in TNG and DS9 she played Lwaxana Troi (mother of Deanna Troi). She also plays Christine Chapel in some Star Trek movies.
