Hank Azariah (Moe, Apu) has been on several other shows, most notably as Nat the dog walker on "Mad About You".
Dan Castelanetta (Homer) has appeared on a couple other shows as a guest.

Julie Kavner (Marge) played the sister Brenda on the sitcom "Rhoda", which was a spinoff from the Mary Tyler Moore Show. Also appeared & starred in a few movies, including Hannah & her sisters.
Nancy Cartwright (Bart and others) has quite a list of voices she's done for projects other than the Simpsons, also appeared in a few movies.

Yeardley Smith (Lisa) used to play on Herman's Head. I can't remember her character's name though.

Many of them, including Dan, Julie, Nancy and Yeardley, did sketches on the Tracy Ullman show that spawned the Simpsons in the first place.
I think Dan Castelanetta played a man in a Homer costume in a memorable episode of LA Law where he was being prosecuted for behaving in a drunk, disorderly and generally Homer-like manner.
Apart from "The Simpsons", I know Nancy's voice best as Mindy in the entertaining Buttons/Mindy section of "Animaniacs".

Yeardley Smith who does Lisa's voice has been in the Jack Nicholson film 'As Good As It Gets.'

Yeardley Smith was also on Dharma and Greg.
She played Greg's secretary, Marlene.
