No, the Prince of Wales is the eldest son of the reigning monarch, if they have a son. Girls can only be the Princess of Wales if they marry the Prince (and as we have discovered they lose that title if they are divorced from the Prince).

She was formally called 'The Princess Elizabeth', being the daughter of the monarch, and her sister was called 'The Princess Margaret'. Her father, King George VI, was never Prince of Wales as he acceded to the throne because his brother Edward had abdicated. Queen Elizabeth's parents were the Duke and Duchess of York before they became king and queen.

The eldest son of the monarch is not automatically Prince of Wales, either. Charles was created Prince of Wales in 1969, when he was in his early 20's.

In the case of the title "Prince of Wales", the word "prince" means "ruler", not "son of a monarch".

The sovereign's eldest son is not automatically Prince of Wales from birth, but has to be appointed by their parent. The appointment is not the same as an investiture. Not all Princes of Wales have been invested in the position as Prince Charles was 1969, and that isn't necessary to hold the position. In fact Prince Charles was created Prince of Wales eleven years previously, in 1958.
