Would Mike Sarne with Wendy Richard, "Come Outside" count as a duo?
It was #1 in the UK in 1962.

In the UK, the first male-female duo to have a #1 in the singles chart was "I'm Walking Behind You" by Eddie Fisher featuring Sally Sweetland but that was in 1953.
Next, as solarbears says, was "Come Outside" in 1962 which is credited to both Mike and Wendy although all Wendy did on the record was make comments like "Oi, leave it out".
Soonny & Cher were next with "I Got You Babe" (1965).
Surprisingly, there were only three more such duets to top the UK chart in the 1960s.
These were Frank & Nancy's "Something Stupid" (1967), Esther & Abi Ofarim's "Cinderella Rockafella" (1968) and the breathy "Je T'Aime...Moi Non Plus" by Jane Birkin & Serge Gainsboourg
Of these, only the Sinatras also topped the US chart.
