When you say 'original' do you mean under Captain Pike, in the Pilot Episode? If so, the answer is 203. But if you mean under Captain Kirk, the answer is 428.

To expound upon the above post:
Year 2254 -- Enterprise (under Pike) discovers Talosians on Talos 4 (Enterprise Crew size now - 203).
Year 2267 -- Kirk assumes command of Enterprise (Enterprise Crew size - 428).
