The answer is actually straight down, rather than up. While a river might flow upstream in mad-made situations, at any point where water is going straight down it is considered a waterfall, not a river.

Nile flows North. Many do!
As for straight down.....There are no waterfalls without rivers (bar glacial melt zones) so a waterfall IS part of a River and therefore rivers do, at points, flow straight down!
