Answer: Hand of God

Sam discovered references to the "Hand of God" while researching Nazi history. He discovered they had an archeological branch that had searched for this super weapon that would be strong enough to win the war. "The Hand of God" was a "catch-all" for items God touched while on earth.

The items were said to hold traces of God's power. Sam convinced Dean they needed to search for one of these weapons, lost during WWII, so they could use it against Amara. Once Dean found it, the artifact turned out to be a piece of the Ark of the Covenant.
2. When Dean proposed that Sam stay behind while he and Cas time travelled to retrieve the weapon in "The Vessel," Sam asked, "So you expect me to sit here and ride the pine while you and Cas play _______"?

Answer: Jules Verne

Sam referred to Jules Verne, author of numerous adventure stories, including "Twenty Thousand Leagues Under the Sea." This was a particularly appropriate reference, since Dean was proposing a trip back to the USS Bluefin, a WWII submarine, to retrieve the artifact.

Cas, though, was briefly able to take control in order to save his friend.

They eventually discovered that a demon was collecting souls, and using one of the old wrestlers to kill people. The story of the old wrestler, who had been a hero of the brothers and then sold his soul for one more chance at glory, was poignant.

When Lucifer allowed Crowley to escape so he could catch Crowley trying to retrieve the Rod of Aaron, a "Hand of God," he took time to taunt the former King of Hell, telling him "every demon in Hell" hated him, and capping it off with the crack about Crowley's sort-of friendship with the elder Winchester.

A soul eater was an undead creature that fed on souls. Rufus bet Bobby a bottle of Johnny Walker that it was a Baku, a dream eater. He was wrong, and paid up at the end of the episode. That was the same bottle that Jodie found when she and Sam were going through Bobby's boxes after his death in "Time after Time after Time" in season 7.

When a soul eater "yanks a person's soul" into their nest, the person is in a place that "exists outside of time and space," according to Rufus. So, both Bobby and Dean were in the nest at the same time. And, while it was never explained, the viewer could infer that Dean saved Bobby back in season 4 or 5 by killing the soul eater and releasing him from the nest in season 11. (Like Sam, this makes my head hurt; and like Dean, I need a drink.) Dean also saw Sam in the nest, but that was because the soul eater "messes" with its victims by showing them people they care about, and showing them dead. Mary Henderson from Bobby's case saw her husband in the nest, and Bobby saw dead Sam and Dean.

Sam was shot, and after being suffocated, appeared to die. It turned out he was just in shock. Dean swallowed a pile of pills so he could die and make a deal with a reaper to save his brother - similar to what he did in "Appointment in Samarra," in season 6, when he had Dr. Roberts kill him so he could talk to Death and get him to retrieve Sam's soul from Hell.

This time, the reaper, Billie, told him no deals, but also told him Sam was still alive. And, the doctor was able to bring Dean back.

Answer: Werewolves

The brothers were fighting pureblood werewolves, attempting to save the monsters' victims. They killed two of the werewolves, but Sam was shot by one. Sam later killed two more. One of the "victims" had also been bitten, and turned into a werewolf, too. Sam killed that werewolf when the newly-turned werewolf was attempting to kill Dean after killing the doctor and sheriff.

This same "victim" had also tried to kill Sam.

Dean killed himself in order to bargain with the reaper to bring his brother back. He begged her to save him, tried to convince her that Sam was the only one that could save the world from the Darkness, and then offered to trade his own life for Sam's. Billie refused to bargain, and practically licked her lips at the prospect of reaping Dean. But, then she revealed that Sam was still alive, telling his brother, "Trust me. If the Big W bit it, I'd get a call."

Crowley got his hands on another "Hand of God," the Horn of Joshua. (Joshua won the battle of Jericho.) After a lot of haggling about who would use the weapon, and an unsuccessful attempt to expel Lucifer from Cas' vessel, Lucifer tried to use it against Amara.

It did not appear to affect her at all. This left the Winchesters, Crowley, and Lucifer with no plan to stop God's sister from destroying the world.

Rowena explained to Amara that she had placed some powerful magic within her body and when Lucifer killed her, the magic went to work and she was revived. The witch helped Amara to recover from the attempted angel smiting.
