According to Jean-Claude, it is impossible to be marked by more than one vampire at a time. But, of course, it happens to Anita. A side effect of this was that it wiped out all the marks she was carrying from Jean-Claude. So once she survives gutting Alejandro--she is free of all vampire marks.

It is common knowledge that to kill a vampire, it kills the human servant also and vice-versa. But Anita survives, because she is one tough cookie.

9. How does Anita find out that Richard is a werewolf?

Answer: He gets his throat ripped out and survives.

Since Richard is alpha, he is very good at hiding what he is. Since people tend to die on Anita all the time, she considers that it may be a benefit that only silver can kill him.

10. Why does Jean-Claude give Anita the third mark?

Answer: She's dying from Lamia poison.

Anita is furious and scared. It is what prompts her into betraying Jean-Claude to Mr. Oliver. Anita would rather die than be bound to Jean-Claude. Richard was the one who brought her to Jean-Claude for him to save her. The third mark is where the vampire drinks the blood of the servant. Once Anita realizes that Mr. Oliver is planning a bloodbath as soon as he has control over the city vampires, Anita allies herself with Jean-Claude again.
