Most of the season takes place on the Greene family farm, which serves as a base location for the group's search for Sophia. The farm has been in Hershel Greene's family for over 100 years and was relatively untouched by walkers. He lives there with his daughters Maggie and Beth, Beth's boyfriend Jimmy, and his farm hand Otis and Otis's wife Patricia.

Farmhands Otis and his wife Patricia also die in season 2, as do Tony and several members of his gang: Dave, Sean, and Randall. Embarrassed and fearing he'll get in trouble, he doesn't tell anyone what happened. Later, the walker attacks Dale while he's on patrol and disembowels him.

In episode 1 of season 2, "What Lies Ahead," Carl is shot while gazing at a deer. In episode 4, Maggie and Glenn have sex at a pharmacy where Glenn is retrieving a pregnancy test for Lori, and Lori discovers she pregnant. Daryl finds Sophia's doll in episode 5.

The Trail of Tears is the name given to the forced relocations of tens of thousands of Native Americans from their homelands after the Indian Removal Act of 1830. Daryl tells Carol how while the Native Americans were making the journey from their homes, the mothers wept for their children who were dying of starvation and exhaustion along the way. "So the elders," Daryl tells Carol, "they said a prayer; asked for a sign to uplift the mothers' spirits, give them strength and hope.

The next day this rose started to grow where the mothers' tears fell. I'm not fool enough to think there's any flowers blooming for my brother.

But I believe this one bloomed for your little girl."

They take him back to the farm to treat him and decide what to do with him.

Shane secretly retrieves Randall from the barn, leads him out, and snaps his neck. He then bangs his own head on a tree to make it look like Randall assaulted him to escape. He hopes while the group is out searching for Randall, he can kill Rick. Daryl and Glenn find Randall's walker form and are confused as to why it has no bite marks. (They do not yet know one can turn after dying without being bit.) Shane also kills Otis, in an earlier episode, by shooting him and leaving him to be devoured by walkers so he can get away.

Andrea contemplated suicide at the CDC, but Dale forced her out of the situation. Beth is distressed at having seen the walkers in the barn and because her walker-mother almost bit her. Beth is prevented from her suicide attempt by Maggie and Lori, though Andrea later leaves her unguarded to choose whether or not to try again. Beth locks herself in a bathroom, shatters the mirror, and uses a shard to cut her wrists, but she only makes a shallow impression, and Andrea smiles that she chose to live.

Daryl later throws Beth's suicide attempt back at her in the moonshiner's cabin where they play "I Never." Angered at her assumptions about him having spent time in jail, they fight, and he says, "I sure as *** never cut my wrists looking for attention."
