After you finish the Enigma side mission, you can go to the Batcave and Alfred reveals his real name. The other aliases are used in other variations of Riddler. Edward Nigma is his most common AKA and was used in "Arkham City". Coleman Reese was in "The Dark Knight", so when when put the Mr. in it, it sounds like mysteries. Sean Shivell was an AKA used in the comics.

The poison is the after effects of Joker's Titan overdose, back in the first game "Batman Arkham Asylum".

This quirk also makes scoring points on Stage 5 particularly difficult on the Mania setting, and for that reason, attempts to score a record number of points are best attempted on the Easy and Normal settings.

The Riddler's challenges include everything from question mark-shaped Riddler trophies, Interview Tapes, and chattering teeth. Each of these will give Batman more experience points. Aside from receiving trophies/achievements for collecting certain percentages of materials, it can be a real reward for the ultimate completionist to find everything.
