Jack Lemmon was the first actor and Helen Hayes was the first actress.
Helen Hayes The Sin of Madelon Claudet (1931) Airport (1970)

Jack Lemmon Save the Tiger (1973) Mister Roberts (1955)

On an interesting note, Barry Fitzgerald became the first (and only) actor to be nominated in two categories (Best Actor and Best Supporting Actor) for the same role in the same movie (he won Best Supporting Actor). The Academy later changed the rules which prevented this in future nominations.
