After being injured in an automobile accident, Duncan Marsh, an amnesiac man, is treated by Drs. Morgan Chambers and Emery Forrest. Dr. Chambers is fearful that radioactive fallout from nuclear bombs will destroy humanity. He is working on a serum, derived from wolves, which he intends to use on himself and a few selected humans, to survive.

The unscrupulous doctors inject Marsh with "irradiated wolf serum" as an experiment. A side-effect of the serum is that Marsh turns into a werewolf when under emotional stress.

Toho Films was asked by AB-PT Pictures to make a monster movie for television. The script anticipated three thirty-minute episodes with commercial breaks built in. The aspect ratio was 1.33:1. AB-PT withdrew from the partnership while the movie was being produced. Toho changed course to produce a theatrical film instead.

The Toho Pan Scope aspect ratio was 2.35:1. Editors were forced to "blow up" and crop the film already shot to fit the widescreen format. The Japanese version was released and then heavily edited with new American cast members and extensive new footage for English-speaking audiences.

Dr. Abel is conducting research on keeping tissues alive until transplants can be obtained. He develops Serum Z which is what keeps the dog's head alive, after it has been removed from its body. Dr. Abel has a heart attack while performing surgery and his colleague Dr. Odd keeps his head alive by using the serum.
