1. Which character, leader of the Turkish Assassins, greets Ezio when he first arrives in Constantinople?

Answer: Yusuf Tazim

Upon arrival, Yusuf Tazim gives Ezio a brief rundown of Constantinople and the current state of affairs as they make their way through the streets of the Galata district to the Assassin hideout in the northern section. Yusuf equips Ezio with a hookblade and teaches him how to use it effectively in combat and to facilitate the climbing of buildings. Yusuf proves to be a valuable ally, and aids and accompanies Ezio on many of his missions. Ultimately, he is slain by Templars whilst trying to protect Sofia Sartor from attackers.
2. In the Mediterranean Defence mini-game, which city is unlocked after playing specific multiplayer maps?

Answer: Rhodes

After completing a round on the Ippokratous, Souk and Knight's Hospital maps in the game's multiplayer mode, Ezio can begin to send his assassins on contracts in Rhodes. The Mediterranean Defence game functions much the same as the assassination contracts from 'Assassin's Creed: Brotherhood', insofar as Ezio can assign his assassins to complete various tasks throughout the Mediterranean. Quite a few of these tasks involve reclaiming the respective city from the clutches of the Templars, or defending it if it is under attack. If the Assassins can control all cities, excluding Rhodes, at once, then an achievement/trophy will be unlocked.
3. In which district of Constantinople is Sofia Sartor's bookshop?

Answer: Imperial

As Ezio searches Constantinople for clues leading to the whereabouts of more Masyaf keys, he finds himself investigating the old trading post of Niccolò Polo, now a bookshop run by Sofia Sartor, a Turkish literary scholar who had returned from Venice where she grew up.

The pair bond instantly, and Sofia later helps Ezio in his quest by locating and translating old books. Just as Ezio is leaving Cappadocia, he receives news that Sofia has been apprehended by Templar agents, and manages to save her before she is killed by a Byzantine. Though not explicitly stated in the game, but apparent in the animated short film 'Assassin's Creed: Embers', Sofia and Ezio get married, and live together in Tuscany with their two children, Flavia and Marcello.
4. Piri Reis, an Ottoman Assassin, specialises in what?

Answer: Bomb crafting

Piri Reis provides Ezio with the fundamentals of bomb crafting, a valuable technique which can make his work as an Assassin just that bit easier. The game's crafting mechanics involve selecting three components to make a bomb: the shell, the gunpowder, and the active ingredient.

The bombs can then be placed into one of three categories - lethal bombs that harm enemies, tactical bombs which offer Ezio a temporary advantage over his foes by incapacitating them briefly, and diversion bombs that draw enemies to or away from a particular area so that Ezio can either sneak past or take them all out at once with more bombs or attacks. Crafting ingredients can be obtained throughout the city by opening treasure chests, looting dead bodies and purchasing from black market dealers, among other things.
5. Ezio gains a valuable ally in the Prince of the Ottoman Empire. What is his name?

Answer: Suleiman

Suleiman and Ezio first met aboard the ship when the latter was just arriving in Constantinople. Suleiman told Ezio that he was just a student, but Ezio finds out later that he is the Prince when he saves him from a Byzantine attack at his residence at Topkapi Palace. With Suleiman's allegiance, Ezio is able to infiltrate the Janissary order and gain vital information about the current political climate in Constantinople.
6. Desmond Miles also faces trials of his own whilst stuck on Animus Island. How many 'Desmond sequences' are there?

Answer: Five

As Ezio travels around Constantinople and Cappadocia, he collects Animus Data Fragments scattered all over the place. These fragments unlock special sequences for Desmond on Animus Island. The trials see Desmond, in a first-person view, working his way through a series of strange rooms, with only the ability to spawn floating blocks to help him.

At several points throughout these sequences, more of Desmond's back story is revealed as the Animus taps into his memory and pieces of his subconscious try to fit themselves together.
7. How many Masyaf Keys, needed to unlock Altaïr's library, is Ezio searching for?

Answer: Five

The first part of the game takes place in Masyaf, and Ezio fights Leandros, a Templar captain, for possession of the journal of Niccolò Polo, which reveals the locations of the five Masyaf Keys. Each key is encoded, if you like, with a memory of Altaïr which Ezio relives. Four of the keys are hidden in Assassin tombs around Constantinople, but the last is in the possession of the Templars, and Ezio hunts down and kills the man who holds it, Manuel Palaiologos, in Cappadocia.

Inside the library, Ezio finds the skeleton of Altaïr himself with one final key and one final memory. Shortly afterwards, Ezio relinquishes his life as an Assassin and returns to Constantinople with Sofia.
8. Who addresses Desmond when he achieves a synch nexus inside the Animus?

Answer: Jupiter

When Desmond meets Subject 16 inside the Animus, Sixteen advises that Desmond achieves a synch nexus if he wants to exit the Animus. A synch nexus occurs when all the memories have been relived and there are no more left, at which point Ezio's and Altaïr's memories separate from Desmond's and he enters the Nexus of Time where Jupiter, one of the First Civilisation, addresses him. Jupiter tells Desmond where the location of the Grand Temple is, so that he may journey there and find a way to prevent or safeguard the planet against the catastrophic solar flare that threatens the Earth.
9. Ishak Pasha's tomb can be found inside which Constantinople landmark?

Answer: Hagia Sophia

The iconic Hagia Sophia houses many secrets, but none more precious to Ezio than the tomb of the former leader of the Turkish Assassins, Ishak Pasha. Scattered across the city are ten pages from Ishak Pasha's memoirs that reveal, when all have been collected, the location of the tomb, and subsequently, the location of Ishak Pasha's armour.

This unbreakable armour has the same resistance and health bonus as the Master Assassin armour, so it basically comes down to cosmetics when choosing between these armour sets for Ezio.
10. The multiplayer mode returns from 'Assassin's Creed: Brotherhood', with some new additions and improved mechanics. Which of these is not one of the four stages of the Approach Meter?

Answer: Unnoticed

When searching for your target, your Approach Meter stays at Discreet. When your target enters your line of sight, the meter begins to increase to the next threshold, Silent, as long as you give yourself away by performing High Profile actions. The meter can then increase further to the highest threshold, Incognito. Should you be Discreet and perform a High Profile action, the Approach Meter will start to decrease down towards Reckless as long as you don't start a chase.

A Reckless kill yields +50 points towards your kill score, Discreet gives +100, Silent awards you +200, and an Incognito kill will result in a massive +350 points.

It pays to be inconspicuous, so be sure to use blending, disguises, and the environment to your advantage to accumulate big points.
